Shirley Bousquet a suivi une formation de théâtre, de danse (modern-jazz) et de chant ainsi qu'un BTS Force de Vente au GRETA à l'École Nationale de Commerce de Paris XVIIe.
Son amour pour la comédie est venu très tôt. Vers l'âge de 6 ans déjà, entre copines, elle interprétait des sketchs tous les samedis après-midi. Sa mère rejoignait tous les weekends son père, skipper, à Saint-Malo ; ayant peur de la route, elle lui enregistrait des sketchs sur cassettes pour être sûre qu'elle ne s'endorme pas au volant.
Shirley Bousquet a commencé sa carrière professionnelle à 15 ans au Bataclan avec la Compagnie des Saltimbranques avant de rejoindre la Ligue d'improvisation.
C'est grâce à la télévision qu'elle acquiert sa notoriété. Tout d'abord avec son personnage de Nancy dans la série Caméra Café qui la révèle au grand public, puis sur Canal+ dans l'émission de Stéphane Bern, la Bern Académy où elle interprète des sketchs avec Florence Foresti alors débutante. C'est tout naturellement qu'elle reprend son personnage au cinéma dans l'adaptation de la série Espace détente (2005). Elle tenait également le rôle de Jeanne dans Sous le soleil.
