After graduation, from 2001 to 2003, she participated in several theater productions. She later moved to Milan to pursue a modeling career. She posed for a Brooksfield photo campaign, walked the runway for Il Marchese Coccapani, for Max Mara and for Carlo Pignatelli.
During the same period, she also appeared in several commercials and two music videos, Tu no by Gemelli DiVersi and Che tempo fa by Miotti. In 2005, she returned to Rome, joining Rai as a correspondent on the program Guarda che luna, hosted by Massimo Giletti and Hoara Borselli. In the same year, she hosted the program Italia che vai with Guido Barlozzetti on Rai 1. The following year, she hosted the program Caccia al ladro, broadcast by Sky's Leonardo channel and rebroadcast, in 2008, by the Facile TV channel on digital terrestrial. In 2007, she hosted the Castrocaro Festival together with Massimo Giletti, while on December 15 of the same year she presented the Rai 1 afternoon program Effetto Sabato.
In the summer of 2008, she was the host of Sabato & Domenica Estate, a weekend program on the air in the morning from 6:50 to 9:30 a.m. with Attilio Romita, replacing Sonia Grey and Franco Di Mare, who returned in the 2008/2009 season with Vira Carbone, and of Pongo and Peggy - Gli animali del cuore, aired on Saturdays at 11:45 a.m., also on Rai 1, a second edition of which she hosted the following summer. Again on the same network, on July 19, he hosted a zero episode of the program Parenti Talenti. In the summer of the same year, she hosted the late-night program Oltremoda reloaded, on Rai 1. Since December 15, 2008, she has been in charge of Rai 1's well-known midday program La prova del cuoco, replacing Antonella Clerici, who had to leave the program to carry her pregnancy to term. At first, the rotation was supposed to last until the end of the television season, but, in April, the then director of Rai 1, Fabrizio Del Noce, confirmed her at the helm of the program for the following season as well.
In June 2009, she also took over from Antonella Clerici in hosting the Wine Oscar and, also in the summer, she hosted the Castrocaro Festival event on Rai 1. Since mid-November 2009, she has conducted, on Rai Radio 2 the program Le colonne d'Ercole together with Armando Traverso and Federico Biagione.
On January 6, 2010, Isoardi was on the link from the State Monopolies for the program Affari tuoi on Rai 1 combined with the Italian Lottery. In June 2010, she was again the host of the Wine Oscar. Since September 2010, following Antonella Clerici's return to La prova del cuoco, she has been entrusted with the hosting of the Sunday program Linea verde. Starting October 9, 2010, she also hosts the Rai 1 program A come animali, aired on Saturday mornings. Together with Pupo, she hosted Miss Italy in the World on July 4, 2011.
From Aug. 6 to Sept. 11, 2011, he hosts on Rai Radio 2 I miei e i tuoi with Gianfranco Monti. On August 9, he presented on Rai 1 the "Persephone 2011" award dedicated to theater. Since September 12, 2011, he has been hosting Unomattina together with TG1 journalist Franco Di Mare. On December 25, 2011, he hosted Canto di Natale on Rai 1. On May 9 and 16, 2012, he hosts Punto su di te! on Rai 1 with Claudio Lippi. Since September 2012, and with Franco Di Mare of Unomattina she also hosts Unomattina Rosa. In 2013, she was confirmed as the host of Unomattina together with Duilio Giammaria with the participation of Eleonora Daniele and Massimiliano Ossini.
In the summer of 2014, she hosts on Rai 1 the musical event Una notte per Caruso together with Flavio Montrucchio. She began the 2014/2015 television season on Rai 1 as the sole host of the daily program A conti fatti. On Saturday, July 4, 2015, flanked by Flavio Montrucchio, she hosts the event Una notte per Caruso in prime time on Rai 1 for the second consecutive year.
In 2016 she appears on the cover of the album Musica Endemica by the Occitan group Lou Dalfin.
In the 2018/2019 television season, he returns to host La prova del cuoco on Rai 1.
When she finished hosting La prova del cuoco with the program's closure in June 2020 due to an obvious drop in ratings (as Rai 1 director Stefano Coletta declared), in September of the same year she participated as a contestant in the 15th edition of Ballando con le stelle paired with dancer Raimondo Todaro. She withdrew in the seventh episode due to a sprained ankle, but, thanks to repechage, managed to make it to the finals.
A few months after her adventure in the Rai 1 dance show, she participated as a contestant in the fifteenth edition of L'isola dei famosi, on Channel 5; as happened in Milly Carlucci's program, she had an eye injury, initially not worrying, later led to her withdrawal from the game.
